About ACE-031

ACE-031 is not a peptide but a fusion protein: the extracellular, ligand-binding part of human activin receptor type IIB (ActRIIB) joined to the Fc region of human IgG1. It works as a decoy receptor, binding myostatin, activin A and other TGF-β family ligands that would otherwise signal through ActRIIB. Its size shows in the table: 77489.89 Da.

Its clinical development was stopped. The published record includes a first study in healthy volunteers (Attie et al., 2013) and a 2021 review by Lee of approaches that target the myostatin pathway, and a 2025 study describes how ACE-031 offered on the black market can be detected by gel electrophoresis (Reichel et al.). ACE-031 is listed under Hormone & growth axis.

Why does ACE-031 have a different purity line from the peptides?

It is a large recombinant protein, so its purity is measured by size-exclusion and reversed-phase HPLC and specified at ≥95% (SEC/RP-HPLC), as the table shows.

Is ACE-031 an antibody?

No. It contains the Fc part of an antibody, but its binding part is a receptor domain, not antibody variable regions, and it binds several ligands of the ActRIIB pathway rather than a single target.
