About AICAR

AICAR is not a peptide. It is acadesine, the riboside of 5-aminoimidazole-4-carboxamide — a small nucleoside that cells convert to ZMP. Because ZMP resembles AMP, AICAR became the standard laboratory tool for switching on AMP-activated protein kinase (AMPK), the enzyme cells use to sense a shortage of energy.

It is listed under Longevity & mitochondria with the other cell-energy compounds. The study cited most often is the 2008 paper in Cell by Narkar and colleagues on AMPK activation and exercise-like gene programs in muscle. A 2021 systematic review by Višnjić et al. collects the effects of AICAR that do not run through AMPK, which matters when you interpret results. Acadesine also went through clinical trials (Mangano, 1997) and is not an approved medicine.

Does AICAR act only through AMPK?

No, and the literature says so: the 2021 review by Višnjić et al. lists effects that are independent of AMPK. Plan controls that can separate the two kinds of effect.
