About IGF-1 LR3

IGF-1 LR3 is an engineered form of insulin-like growth factor I. Compared with the native hormone it carries an N-terminal extension and an arginine substitution — the R3 in its name — for 83 residues in all, held together by three disulfide bonds. Both changes weaken binding to the IGF-binding proteins, which otherwise hold native IGF-I back in culture media and in serum.

The analogue was described by Francis and colleagues in 1992 and is widely used in cell culture. Research on it and on IGF-I more broadly covers satellite cells and muscle growth (Barton-Davis et al., 1999) and the signalling behind muscle hypertrophy and atrophy (Yoshida et al., 2020).
