About Melanotan I

Melanotan I is afamelanotide: α-MSH with two changes — norleucine in place of methionine and D-phenylalanine in place of the natural L-form — that make it resistant to proteases and more selective for the melanocortin 1 receptor (MC1R) on pigment cells. It is linear, unlike the cyclic Melanotan II.

The first human work on it measured skin pigmentation (Levine et al., 1991). As afamelanotide in an implant formulation it later became an approved medicine for a rare light-sensitivity disorder (Langendonk et al., 2015); that approval does not cover this research powder. It sits in Skin, hair & pigment.

Melanotan I. Its receptor activity centres on MC1R, while Melanotan II acts on several melanocortin receptors. The two also differ in structure (linear versus cyclic), CAS number and molecular weight.
