About PNC-27

PNC-27 is a hybrid of 32 residues. Its first part is residues 12–26 of the tumour-suppressor protein p53, the stretch that binds HDM-2 (the human counterpart of MDM2); its second part is a membrane-residency sequence that holds the molecule at the cell membrane. The full sequence, formula and molecular weight are in the parameter table above.

Most published work on PNC-27 comes from the Pincus and Michl laboratories and concerns how it binds HDM-2 sitting in the membranes of tumour cells and the membrane pores that follow (Sarafraz-Yazdi et al., 2010 and 2022), with a 2024 paper looking at mitochondrial membranes as well (Krzesaj et al.). It is a research peptide with no approved use, listed under Longevity & mitochondria.

Is PNC-27 the same as a p53 peptide?

No. It contains the p53 segment (residues 12–26) but adds a second sequence for membrane residency, so its mass, formula and behaviour differ from the p53 fragment on its own.

What does the COA confirm for a hybrid peptide like PNC-27?

HPLC purity shows how much of the lot is the main product, and MS identity checks that the main product has the expected mass for the full chain (4032 g/mol in the table above). Together they screen for shortened chains left over from synthesis.
