Retatrutide is a single peptide that activates three receptors: GIP, GLP-1 and glucagon. It was first described under the code LY3437943, from discovery through early clinical work (Coskun et al., 2022). Compared with Tirzepatide, which acts on GIP and GLP-1, it adds the glucagon receptor; compared with Mazdutide and Survodutide, which act on GLP-1 and glucagon, it adds GIP.
